Which objective must be in position when you first set up a microscope?

I have always had the longest objective in position (pointing down), then lowered the lens to what I thought would be close to a good focus. This way, you can rotate the turret to the shorter lenses, and be assured that the lens body will not strike the stage or slide.

What are the steps to focus a microscope?

Depending on the microscope the steps differ but assuming you are using a light microscope (the only microscope you will use before university). The microscope must be set to the lowest magnification with the stage set as far way as possible (the platform the sample is held on). Then using the coarse focus (the larger knob) bring the stage towards the lens while looking though the eye piece when the sample comes into focus use the fine focus knob (the smaller knob) to bring the image to optimum focus. When this is achieved increases the magnification (by changing to a stronger lens) and using fine focus continue to focus the image and keep repeating the steps till a max focus is achieved. If more information is need let me know as there are more complicated techniques but this should be enough for before a university level depending on what country you are in.

What are the 2 basic principles involved in the use of the microscope?

It's important to hold the microscope set up properly so exacting results can be obtained while in employment. Make certain both the capacitor as well as any field diaphragms is completely exposed while in usage. If this is not performed properly, resolution will be greatly diminished. Make sure the illumination system is set up to full brightness and the blue filter is used. Diminished illumination will affect resolution and degrade it.

Why do you place the microscope at least one inch from the egde of the table?

Microscopes are very valuable pieces of equipment. This set distance is a simple safety precaution. Also, if you have spent a long time preparing the sample on the slide, the very last thing you want to do is knock over the microscope and contaminate the slide!

What part of the microscope supports the body tube which connects the eyepiece to the set of objective lenses?

Make Sure To Open And Close?

Slide in campers are great if you want to save room. When you set up the camper, you have to remember to slide out the part of the camper that you will use. This part is usually either the sleeping area or part of the living area. You can slide the camper out when you want to stay in it at night, or you can keep it out the entire time you are camping. After you slide the camper out, you need to remember to slide it back in before you leave or it could damage the camper or the vehicle.

Why do you have to watch from the side when changing objectives of the microscope?

Sometimes, if the stage is set high enough, the objective lens can hit or scratch the slide when it's changed, because each objective lens is a different length. From the back of the microscope, you cannot see if the lens is going to hit the slide or not, and run the risk of disturbing your sample. This problem can be prevented by looking from the side and stopping if it looks like the lens is going to hit the slide.

How do you look for vacuole at onion cell through light microscope?

You should use iodine to stain the cells. Set the cells on a slide, let them dry you will repeat the process until it is completely covered in iodine and then you will slide it in the stage and then hopefully you will then see the sells of the onion skins.

Why should you always view a slide at a power first?

When viewing specimens or objects under a microscope, it should be set to a low power first. This allows for a wider field of view that makes an object easier to locate.

Why should you watch from the side before you lower the objective of your microscope?

Sometimes, if the stage is set high enough, the objective lens can hit or scratch the slide when it's changed, because each objective lens is a different length. From the back of the microscope, you cannot see if the lens is going to hit the slide or not, and run the risk of disturbing your sample. This problem can be prevented by looking from the side and stopping if it looks like the lens is going to hit the slide. Watching from the side also helps to know if it is too low, or just in the right distance.
