There is no nationwide ranking of the "highest" law enforcement agency in the country. All agencies tend to their own specific areas of jurisdiction and/or expertise. (e.g.: the FBI doesn't 'out-rank' the BATF or the ICE or the Tresuary Department or the Secret Service, any more than the LAPD 'outranks' the NYPD, the MPDC, or the Pinellas County Sheriffs Office). All are established to fulfill certain enforcement areas and operate within those parameters. Some have an expertise which other departments or agencies may call on and rely (Treasury for counterfeitting - BATF for firearms - FBI for laboratory facilities - etc) but all operate pretty much independently of one another as they pursue their own areas of assignment.

It depends on the crime that's being investigated. From a State constitutional point of view the Sheriff Office will be the highest Law Enforcement agency within their respective counties.
