I think the seeds of the American Civil War were planted when England formed the Thirteen Colonies in the 17th Century. The Northern colonies were founded by religious separatists (Pilgrims, Puritans, Quakers, and others who couldn't hang with the Church of England), while the Southern colonies were less religious, more minded toward expansionism and profit, and had fewer scruples about human slavery. Since you asked, I think the North and South were two separate countries long before they became one country, split into two countries, and became one country again.
