Humans may be able to survive, but many billions could die off from starvation due to lack of food because ash would block out the sun. However, Yellowstone lies on a hot spot and erupts every 600000 years. The last eruption was 640000 years ago, meaning either were overdue or Yellowstone moved past the hot spot and is unable to erupt.

A 'nuclear winter' would ensure, generated by the amount of ashes thrown into the air and preventing sunlight to reach the earth. The size of the explosion would be directly relative to the length of the 'nuclear winter'.

If the volcano under Yellow Stone National Park were to erupt, a wide radius would be under immediate danger from the explosion and lava released. However, the eruption would release millions of tons of ash into the air, which would be carried by the wind and would be enough to circle the entire world. This would block out the sunlight long enough to trigger a new ice age.
