That would probably be "Bullitt," which is a crime drama. McQueen is driving a fastback Mustang. He is stalked by bad guys in a black 1970 Challenger, but he turns the table and chases them in what is considered the greatest car chases of all time. The full chase sequence is here:

McQueen was also in a race movie called, "Le Mans." In this sports car race in France he was portrayed driving a Porche 917K. Although McQueen did not drive in the actual race, a race driver hired by the film drove a camera equipped car which finished something like 2nd.
