Hemoglobin S. This the predominant hemoglobin in people with sickle cell disease. The alpha chain is normal. The disease-producing mutation exists in the beta chain, giving the molecule the structure, a2bS2. People who have one sickle mutant gene and one normal beta gene have sickle cell trait which is benign.

Sickle cell anaemia is such a genetic disease. The sixth amino acid in the beta chain of haeme which is glutamic acid in normal people gets replaced by valine in sickle cell anaemia patients. This simple change in the amino acid sequence results in the the sickle shape of RBCs
The defect is that hemoglobin (the protein) elongates. Instead of being circular, it acquires a crescent shape. With this shape, its harder for it to absorb and release oxygen. Also, when the blood cell reaches the capillaries, it can't fit through as well as normal blood cells can, which is why Sickle-Cell is painful.
