Once you've established your home territory, you'll need to go around the parameter (shown as yellow wedges on the minimap) and press 'p' to mark by urination. Once your territory as at ~95%, you'll progress to the next mission.
Howling without stopping the animation by way of walking or performing another action will also give your territory a small marking boost as you as you allow it to finish.

There are four dens in WolfQuest on Slough Creek. One is a rock-like den that's over on Saddle Meadows, one is another rock-like den on Aspen, one is a tree den over by Bison (the place, not the animals, hehehe), the last one (this is the "secret" den) is in the middle of the one near Bison and the river.
