The three holiest cities in Islam are Mecca, Medina, and Jerusalem (in that order).

Mecca is considered holy because it is where Muhammad grew up and had the vision during the Night of Power which founded Islam. It is also where the first community of believers were however they were highly persecuted by the local pagans. Medina is where Muhammad and his followers migrated during the Hijrah and allowed Islam to flourish and spread. Jerusalem, which was conquered by the second caliphate Umar, is now where the Dome of the Rock stands and reminds Muslims of the place where they think Muhammad ascended to Heaven.
