The Civil War, by far. There were more men killed in the Civil War than in all the other wars the US has ever been in put together, or very nearly. The official death toll of the Revolution is only some 4,000. This is a vast, huge understatement of the actual number, counting only those anybody bothered to write down and report. But still it was surely way under 50,000. There were 620,000 dead in the Civil War. Again that's the official total of military deaths only, and is probably a low number, just the best estimate. The population of the US was ten times larger during the Civil War than it had been during the Revolution, but still, proportionately, the Civil War was deadlier.

Of the 620,000 dead in the Civil War, 360,000 were Yankees and 260,000 were Rebels. Disease killed two thirds of them, on both sides.
