The bloodiest day in American history was August 6, 1945 at Hiroshima, Japan. Over 70,000 people died immediately on impact of "Little Boy", the first atom bomb. Nearly twice that number died later due to the after effects. The second bloodiest day was August 9th, 1945 at Nagasaki, Japan, where between 45,000 and 75,000 people were killed. As for American deaths, the bloodiest day was September 17, 1862 at Antietam - 3,654 deaths, over 23,000 casualties. 9/11 is second in line. However, 100,000 Japanese were killed in the firebombing of Tokyo in World War 2.

About 3,500 people died in the Sept 11 attacks, outnumbering the deaths from Pearl Harbor (2403).
Over 8000 people died in the battle of Gettysburg which took about 3 days. It might have had the most people killed in the US in one day, but it is hard to tell, as the majority of the 46,286 casualties were injuries.
On D-Day, approximately 6,600 US soldiers became casualties (the majority of which were injuries).

Ranged against the migratory tribes of the Cimbri under Boiorix and the Teutoni were two Roman armies, commanded by the proconsul Quintus Servilius Caepio and consul Gnaeus Mallius Maximus. Roman losses are described as being up to 80,000 troops, as well as another 40,000 auxiliary troops (allies) and servants and camp followers --- virtually all of their participants in the battle. All deaths attributed to weapons prior to the invention of gunpowder." Basically, one very large knife fight.
