Eyre's first major journey was his attempt to be the first to overland sheep from just west of Sydney to Adelaide. This journey began on 21 December 1837, departing from Limestone Plains. this particular expedition was unsuccessful.

Eyre's next journey was to explore the countryside beyond the headwaters of the Hutt River, 100km north of Adelaide, which had not yet been settled. this journey commenced on 1 May 1839.

Eyre's expedition to be the first to cross Australia from south to north departed Port Lincoln on 5 August 1839. His journey was thwarted by a series of salt lakes which Eyre believed to be one gigantic horseshoe-shaped inland sea, because by sheer bad luck, he was unable to find any of the breaks between the lakes. This was when he changed his plans and decided to try to be the first to cross from east to west. He returned south to Fowler's Bay and sent some of his men back to Adelaide, opting to take a smaller exploration party across the Nullarbor. This journey departed Fowler's Bay on 25 February 1841.
