About 1839-1840. The author's title page describes the setting as the Mississippi Valley, "forty to fifty years ago." The book was first published (in London) in late 1884 and (in New York) in early 1885. Even without this declaration by the author, we get a good approximation of the year from several details in the story:
1) Huck and Jim believe that Jim, a runaway slave, would be a free man in Illinois; this dates the story prior to the Fugitive Slave Act of 1850.
2) In Chapter XXVI, a servant girl asks Huck for information about King William IV, and Huck the narrator remarks that William IV has been dead for "years." William IV died in 1837.
3) In Chapter XIX, when the two con men calling themselves the "Duke" and the "Dauphin" talk aboard the raft, the "Duke" tells the elderly "Dauphin" that he can't possibly be the long-lost Dauphin of France (Louis XVII, 1785-1795) because he looks far too old. This passage makes sense only if there is a real discrepancy between the fake Dauphin's apparent age and the age that the real Dauphin would be if he were alive. 1840 works; 1850 does not.
After 1841 and before the civil war. Twain would have had vivid boyhood memories of life in rural Missouri from age 6 to 12, when he quit school following his father's death.
