After Sequence 7. Keep in mind that all aqueducts must be repaired to access the last Lair of Romulus.
More Lairs will become available as you progress in the campaign.

No it will not.
He breaks out of the Templar prison where he was captured in the first game and goes to the Assassins hideout and they also have a animus. He uses the life of Ezio Auditore Da Firenze to learn the ways of the assassins.Another person:Desmond Miles became an Assassin simply for being born into the Brotherhood of Assassins, though he claimed not to be an Assassin. Desmond was kidnapped by Templar Warren Vidic and his "Fellow Templar" Lucy Stillman who was actually an Assassin in disguise, while in the first Assassins Creed game, Desmond finds out Warren is a Templar, he then escaped with Lucy into the second game.
Romulus and Remus.
The twin brothers you're referring to are Romulus and Remus. Legend has it that they were abandoned as infants and raised by a she-wolf. As adults, they fought over where to build the city of Rome, leading to Romulus killing Remus and establishing Rome in 753 BC.
forget everything i said. it was all junk.
The best way to increase Ezio's health is to buy armor from blacksmiths. There are 5 armors in the base game and each have different tiers meaning the higher quality armor increases Ezio health more than the lower quality armor. Below I have listed all the armors and when they become available. (Weakest to Strongest) Roman Armor - Full Set available after Sequence 05 - Total Cost 5500 florins Rondelle Armor - Full Set available after sequence 06 - Total Cost 17100 florins Plate Armor - Full Set available after sequence 07 - Total Cost 31600 florins Suesenhofer Armor - Full Set available after sequence 07 and completion of the shop quest "Exotica" - Total Cost 23200 florins Armor of Brutus - Full Set available after completion of all 6 Lairs of Romulus - Total Cost 0 florins (completely free if you don't mind dungeon crawling)
Romulus did not find the city of Rome, he founded it. Remus did not participate in the foundation of Rome. Romulus was the sole founder. Romulus did not build Rome. The founding of Rome does not refer to the founding of the city. It refers to the creation the Roman state. Romulus unified the peoples who lived on the settlements of the hills which were to become the Seven Hills of Rome under his rule. Collectively, these settlements became Rome. Romulus was the first king of Rome as well as the founder of the city.
