By default all the cells in an MS Excel worksheet are locked, although they don't become so until sheet protection is switched on. Select the cell that you wish to be unlocked and either click on the toolbar button that looks like a padlock, or use the Format... Cells menu, Protection tab (clear the check mark). Then go to the Tools... Protection... Protect Worksheet menu, enter a password (optional, but advised) and click OK. Now the only cell that can be edited is the one that was selected when you unlocked it.
