The question is probably unanswerable. A "ghost town" occurs whenever a city or village becomes abandoned, and this has undoubtedly been happening since prehistoric times. Ruins of many prehistoric settlements have been discovered, and it seems likely that the indigenous inhabitants of nearby towns knew about many of them, but since they generally didn't actually write down anything like "7523 BC - Discovered ghost town", we really don't know when they were "found" (or, indeed, if they were ever actually "lost"; the people who moved out when the village was originally abandoned presumably knew all along where their old village was).

Probably the earliest European ghost town in what is now the United States was the Roanoke Colony, which was discovered to be abandoned in 1590.
