There should be red marking on the front of the envelope called a postmark, it should name the city & state of the mail processing center the letter was sent from. This is not necessarily where it was mailed from, just where it was processed.
United States Postal Service can not guarentee standard mail to arrive by any specific time. It is estimated that standard mail will arrive 2-9 days. If you require letter to arrive earlier you can send it Priority Mail, which will arrive within 2 days in most cases.
What shipping service was used? If it was mailed via USPS Express Mail, it will be there the next business day. If it was mailed via USPS Priority Mail, then it will be there in about 2 or 3 business days (though this is NOT guaranteed). Items mailed via USPS First Class typically arrive in 2-5 business days (again, not guaranteed). Parcel Post (and similar services like Media Mail or Library Mail) typically arrive in 5-7 business days.

Depending on where the letter is going, it may arrive on Monday. If you are sending a letter within your town or a neighboring town that shares the same P&DC, then it will probably arrive at its destination on Monday. If it is going to a town served by another P&DC, it will probably arrive Tuesday or Wednesday.
