The Manhattan Bridge is in between the Brooklyn Bridge and the Williamsburg Bridge. All three of these bridges transverse the East River and connect Brooklyn to the East Side of Lower Manhattan.

On the Manhattan side, the Manhattan Bridge is in Chinatown, and on the Brooklyn side, it's in DUMBO (an acronym for Down Under the Manhattan Bridge Overpass) in between Brooklyn Heights, Downtown Brooklyn, and Vinegar Hill.

The entrance is in the City Hall neighborhood, on the east side of City Hall Park. Go up Church Street, turn right onto Vesey Street, and follow Vesey Street one block. When you come to the end of the block, you will reach an intersection: Broadway runs perpendicular to Vesey Street, heading south, and Ann Street and Park Row split off from and run parallel (or, sort of parallel) to Vesey Street.

You need to take the road on the left, which is called Park Row. Park Row leads you onto the bridge. You will be able to tell which street is Park Row, not just because it's on your left, but because Ann Street (on your right) runs in the opposite direction that you're going, and Broadway (further right than Ann Street) runs perpendicular to Vesey Street (away from the bridge).
