It is the first cylinder on the right side.

Hi. I also have a 2001 Kia Sportage with a misfire on my number 1 cylinder. I was looking up some info on the forums, and one person said it's the cylinder cloest to the radiator. I haven't confirmed it yet, but it might just be.
Passenger side front of engine is the #1 plug.
The firing order for an E350 Ford van with a 460 big block is 1, 5, 4, 2, 6, 3, 7, 8. The number one cylinder is on the left as you face the van, on the end of the engine closest to the grill.
The #1 cylinder is the one closest to the front of the vehicle. ( on the passenger side of the engine )
