This is hard to say precisely. However the Minuteman I and II guidance computers stored their software on read only tracks on a harddisk platter, and the Apollo guidance computer stored its software in core rope ROM. But neither of these actually had an operating system, their software ran directly on the hardware with no intermediate layers.

On MS platforms, the system partition is used to hold the boot files. The boot partition holds all the windows operating system files. Leave it to Microsuck to mis-lable the partition hiearchy. The system partition holds what is loaded and executed first after the computer runs through its preliminary BIOS boot sequence. It tells the computer where to start loading the operating system from; the boot partition. The boot partition is where all the program files (thousands of them) needed by the operating system are stored. If the system partition is deleted; the computer will not find the operating system. If the boot partition is deleted, again; the computer will not find the operating system.
If you do not have any protection to your computer, your system applications are at risk: in basic terms, your C:/ where all your critical computer operating files are stored. That is the first location a virus is designed to attack.

The first operating system or software is widely considered to be the program ENIAC, which was created in the 1940s by the University of Pennsylvania's Moore School of Electrical Engineering. ENIAC was the first large-scale electronic general-purpose computer and was used to solve complex calculations for the US military. The program used a variety of different software components, including a control program and an arithmetic unit. ENIAC was an important milestone in the development of software and was the first program to be able to manipulate data stored in memory. It was also the first program to introduce the concept of a stored program, which allowed for faster and more efficient computing.
