It depends on where on 42nd Street you're coming from, but, the N and R trains (of the N-R-Q-W, the yellow line) stop at 28th Street and Broadway.

If you're starting out from West 42nd Street, then you can get the Downtown N or R at 42nd Street-Times Square (at 7th Avenue).

If you're starting out from East 42nd Street, then you can take the S shuttle train (color grey) west across 42nd Street, from 42nd Street-Grand Central Station (at Lexington Avenue) to Times Square. Then transfer at Times Square to the Downtown N or R.

If you're starting out from the middle of 42nd Street, then you can take the Downtown B-D-F-M (the orange line), one stop, from 42nd Street-Bryant Park (at 6th Avenue) to 34th Street-Herald Square. Then transfer at Herald Square to the Downtown N or R, one stop, to 28th Street.
