The address 25 Hudson Street is between Duane and Reade Streets (home of the original location of Duane Reade pharmacies).
To get there from West 51st Street in the Theatre District, take the Downtown 1 train (of the 1-2-3, the red line) from 50th Street and 7th Avenue to Chambers Street.
If you want, you can transfer at 42nd Street (the Times Square Station) for the Downtown 2 or 3 trains. The 2and 3 are express trains. On the 2 and 3trains, it's only 2 stops from 42nd to Chambers Street: 42nd Street ---> 14th Street ---> Chambers Street.
The 1 is a local train, so there are like, over a dozen stops from 42nd to Chambers on the 1. The 2 and 3 don't stop at 50th Street, so you'd have to take the Downtown 1 one stop to 42nd, and then transfer to the Downtown 2 or 3 to Chambers Street.
When you get off the subway, you will be at the 3-way intersection of Chambers, West Broadway, and Hudson. Chambers runs east/west, and West Broadway and Hudson run north/south. Follow Hudson Street Uptown for 1.5 blocks: Chambers Street ---> Reade Street ---> Duane Street.
Your destination (Ninja New York) is pretty much exactly halfway between Reade and Duane Streets, on the left (west) side of the street.
