He lost badly, and by his own admission, did not run a very effective campaign. He returned to the Illinois state senate for another term, and when he ran for the U.S. Senate in 2004, he showed he had learned from his mistakes by winning the senate seat.

It's not so much that they questioned him: it's that he ran a poor campaign against a very popular candidate. It was actually not a state senate race, but a race for the U.S. House of Representatives, in Illinois's 1st District. Mr. Obama, then a state representative, wanted to try to get elected to congress. He ran against incumbent Bobby Rush, and lost badly-- 61% to 30%. It was his first attempt to run for the U.S. congress, and by his own admission, his campaigning skills were not very polished. He never really connected with the voters, and he did not have the high name recognition or popularity that Mr. Rush had. The good news for Mr. Obama, however, was even though he lost (the only time he did lose an election), he learned from his mistakes and ran a much more effective campaign the next time, getting re-elected to the Illinois legislature for a third time, and then getting elected to the U.S. senate in 2004.
Illinois' governor (a Democrat) will choose a replacement for the 2 remaining years in Obama's term. Somewhat similar answer for Joe Biden's Senate seat, except that in Biden's case there are the full 6 years remaining on his term (he was re-elected to the Senate the same day he was elected Vice President). The replacement senator will fill the first 2 years of the term, and then in 2010 a special election will be held to determine who fills the remaining 4 years.

no, every state gets equal represantation in Senate, 2 senators per state. Representation in the House of Representatives is based on population. bigger states have more representatives. the only way for a small state to lose representation would be in the House of Representatives and there would have to be no one living in the state, which is basically impossible.
