First, I think that we should rewrite this quote with better punctuations to make it easier to understand:
"Courage is resistance to fear, mastery of fear - not absence of fear." (Mark Twain)
I think that "Courage is resistance to fear" means that it is sort of an antagonist to fear. That's pretty straight forward...
Then, "mastery of fear - not absence of fear" probably means that someone with courage will still have fear; it doesn't mean that he won't have or feel any fear, however he will use his courage to "master", control, and/or manage the fear and get passed it.

This quote means that courage is not the absence of fear but the ability to face and overcome fear. It is about having the strength and determination to confront your fears and continue forward despite feeling afraid. True courage comes from being able to control and manage your fear, not from never feeling it at all.

The best way to answer this question is to put it into an every day example....... Two guys (A and B) are at a bar and they both see a female celebrity with her girlfriends at the bar. Male A is too itimidated, shy and full of fear to go up to them. He fears rejection. Male B has the same feelings as male A but goes up to the group of girls anyways because he realizes that that his biggest fear (rejection) is not as important as the opportunity to hang out with that female celebrity and maybe even develop a relationship---- something much more important than the simple fear of getting rejected. This is the best you got? How about on 9/11 when all the police, fire, ems personnel were running into the WTC when everyone else is running out? Or how about the soldier who tosses himself on a live grenade to save the lives of his fellow soldiers?
