Wow! That's a long journey! www.varianostravel.com advertise services from Pireaus (Athens) and Limassol but they don't carry cars. There are few scheduled services to Cyprus; cruisecyprus.com list ferries from various locations - including Southampton every Saturday. However, as far as I am aware, all the car ferry services are on freighters and the freight takes priority so days/times are not fixed. The ferry also takes a long time - around 12 days and is far more expensive than flying and hiring a car in Cyprus (as you're having to pay board and lodging for those 12 days).
