This question is probably quite difficult to answer, as there's no set standard by which to measure the characters' powers. The obvious answer would have to be Superman, who has demonstrated many times over why he deserves the title of DC's strongest. But then again, Lex Luthor, his arch nemesis, is quite brilliant and has given him a run for his money, and now with a power suit of his own, the man can go toe to toe with the Kryptonian himself.
If you wanted to measure the strength of a character in terms of ability, then Batman would most likely be the strongest, having gone toe to toe with Darkseid himself and come out as the clear winner. He is also prepared for everything, always having a plan for every scenario that could happen.
In terms of raw strength, few could rival Doomsday, the only one to ever successfully murder Superman. Not only that, but his ability to evolve beyond his foes makes him one deadly opponent as it is astoundingly difficult to defeat him once, much less a second time. We've also seen how, given intelligence, Doomsday can become an even more dangerous enemy.
In terms of both ability and strength, Superman probably takes the top slot. Despite taking care of many problems with his powers alone, he has shown to be just as capable at figuring out solutions to problems that requires more than just super strength or heat vision to solve. He've also gone toe to toe with Doomsday enough times and has even fought and won against Batman when he had all of Superman's powers. The Pre-Crisis Superman is also widely believed to be the most powerful character ever, capable of turning time backward by spinning around a planet. In the Kingdom Come story, Superman is also widely believed to be the most powerful man in existence, to the point where Kryptonite no longer affect him anymore, and is stated to have the power to contain the superhuman war from spreading across Earth. In various stories and Elseworld tales, Superman often takes on the role of being the strongest and most capable as well.

In the Marvel Universe, its easily Shang Shi, the "kung fu master," who is about as strong and skillful in martial arts as Batman's "mystic" Chinese teacher. Apparently he is so good at Kung Fu, he can take on the Hulk. For best sources though, get the Marvel and D.C. encyclopedias, there are new 2008 editions now, with all the latest info. If you want to establish who is the most powerful of this or that, get those encyclopedias, although, for information on cross overs you will have to thumb through the likes of Wizard magazine or like publications. Just go to your local bookstore and "parasite read" a few Wizard mags.
