It was very likely, Jane Seymour. She gave him the son he so desired. She also was agreeable, and was influential in helping Henry mend his estranged and troublesome relationship with his daughter Mary. Jane was thought to have positively swayed Henry into bringing Mary back to court.

Where Anne pushed, Jane would pull; wisely not testing the limits of her power as Queen and her affections from Henry. Jane was key in reuniting Henry with his daugther Mary, whom he had grown extremely distant from during his divorce from her mother. Jane also did what his other wives could not: she produced a living son.
Jane died 12 days after giving birth to the future Edward VI. Henry became extremely depressed and isolated himself well into the next year.
During Henry's final marriage he commissioned a family portrait of himself and his children. Even though he was married to Catherine Parr at the time and Jane had been dead for nearly decade, he commanded that Jane be painted beside him as his "true wife".
