The first woman to run for President was Lucretia Moss in 1848 for the Liberty Party. She ran against Major General Zachary Taylor.

Although she didn't get a lot of votes, the first female candidate for the US Presidency was (I believe) Angela Davis.

It was Victoria Woodhull in 1872 .

If you mean First Lady as in the President's wife, the first woman to do this would be Hillary Clinton, although she ended up not being the Democratic nominee for 2008.
