Towns and cities in Spain follow the same patterns of establishment as the rest of the world.

Wherever settlers can find prime matter, food and water supply, they are bound to settle down. Ofcourse since the world opened to trade, a great city would have to be located near a river, sea or ocean. Its still pretty important today, as the world trade still relies on cargo ships to move merchandise at a reasonable price.
