John Tyler did not have a Vice President because he was originally elected as Vice President and became President when President William Henry Harrison died.

At that point, the 25th Amendment to the Constitution had not yet passed nor been written, so there was no legal procedure to find a replacement Vice President should the Vice President resign or the Vice President take the President's place.
He was the elected vice-president and became president when President Harrison died. Therefore the office of vice-president was vacant. In those days it stayed vacant until the next presidential election.
