it could be either coarse or fine knob. coarse knob is used to focus image at higher magnification, while fine knob is used to focus on lower magnification

The coarse focus on a microscope is provided to move the instrument quickly to where it is nearly in focus. Some microscopes only have a coarse focus. The fine focus is used to move with slower and more precise adjustment to where the instrument is exactly in focus.

a good way that works for me is use the coarse adjustment till you can start to see something then use the fine adjustment to get it into focus.
