Jerusalem is a holy city for the Abrahamic religions: Judaism, Christianity, and Islam. To other faiths it is not a holy place.
Many cities were important in the middle ages for very different reasons. Some cities, such as Constantinople (modern day Istanbul) was important in defending the so-called "Christian" countries from Muslim invaders. Others, such as Paris or London, were important economic centers for their respective countries. Still other cities were (and still are) important religious sites, like Jerusalem and Mecca.
So other people will have religious knowledge.
Jerusalem was important to the Byzantine peoples throughout its long history (from the 4th century AD/CE until 1453) for the same reason that it was important to any Christian society: it was the scene of the crucifixion and resurrection of Jesus Christ, the seminal figure in Christianity. Along with Jerusalem, of course, numerous other sites in the Middle East and elsewhere were valued by Byzantine Christians for faith-related reasons.
Virtually every place mentioned in the old and/or new testaments of the Bible is located either in Israel or in a country that borders it.
Every religious group wants to exercise political control over its holy sites in Jerusalem and does not wish to bequeath them to any other group.
Probably for the same reasons other religious extremists do.
