The Ptolemies were the Greek ruling dynasty of the kingdom of Egypt. They become embroiled with the Roman civil war between Julius Caesar and Pompey. Pompey fled to Egypt after a defeat and Caesar pursued him there. The co-regent of Egypt, Ptolemy XIII, had Pompey murdered and gave his head to Caesar. Caesar was disgusted and sided with co-regent Cleopatra VII. He helped her to win a civil war between the two co-regents. Cleopatra and Caesar became lover. After Caesar's death she became Mark Antony's lover. Mark Antony and Octavian (the future Augustus) came into conflict. The latter used Cleopatra and an excuse to declare war on Egypt and, through this, fight Mark Antony who was living there. Mark Antony and Cleopatra lost the ensuing battle and Octavian annexed Egypt.
There was a second Ptolemy that was important to the Romans too. He was Claudius Ptolemy or Ptolemy of Alexandria. He was a Roman of Greek origin and a mathematician, astronomer and geographer. He is known for his work defining the motion of the planets and the heavens. He was a geographer and map maker which was very important to the Romans.

Cleopatra married both her younger brothers Ptolomy XIII and Ptolomy XiV. Ptolomy XIII died in the war with Caesar and Cleopatra poisoned Ptolomy XIV. She also had a "marriage" with Marc Antony. I put this in quotes because Antony was married to Octavia at the time of his so-called marriage to Cleopatra.
