Most organelles are very small and require a magnification greater than the x1500 available using a light microscope. An electron microscope is required for greater magnification and resolution. Therefore a small area of the slide is avalible.
Some organelles can only be seen with a microscope called "The Electron Microscope".
It isn't possible to see some organelles with a compound microscope because they are too small.

Maybe to have a clearer visual on the plant cells it contains. Basically the light must be able to shine through the object you are viewing and besides when it is not think it wont hold into the microscope clear square thing.
