Curley thinks that Lennie is smiling because of everyone else is bullying him after him and Slim had a disagreement. E.g after Candy got involved in the bullying by saying, "glove fulla vaseline" as an attack towards Curley.

But if that is what you are talking about than it is because George has once again told Lennie the story about their plan for the ranch and given Lennie this wonderful scene to put himself in. He is still laughing about that when Curley walks in and thinks he is laughing at him.
