It represented the first peaceful transfer of power between political parties (in this case the Jeffersonian Republicans displaced the Federalists). It was intended that in a republican society changes in power(technically a revolution) would be determined by the votes of the people as opposed to the old standard of war.
The American Constitution gave the people the power to change all of their leaders simply by voting in an election. In nearly every other country, turning out the leaders required a violent revolt at that time. The elections that we take for granted were a fantastic new thing at the time the Constitution was created.
