The test compound is not necessarily the ultimate carcinogen. In many cases the resulting products in metabolism rather than the compound itself causes cancer. Say, you feed a lab animal with a test compound and it gets cancer. You analyze the animal and you will find that the initial compound is metabolized to other chemicals that directly caused cancer. So this is why a test compound need to be metabolically activated, that is, it has to be metabolized by body enzymes (rat liver has a lot!) and then added to Salmonella to see the mutagenicity.
