- To eliminate hydrogen bonds with sodium hydroxide (NaOH)
- To denature double stranded DNA into single stranded DNA
For neutralization :-
- Neutralize the gel to get the pH that DNA can bind to the membrane.
- Destroy any remaining RNA present in sample
