A 4.3 is essentially a 350 with 2 cylinders cut off. The 4.3 belhousing will bolt to both small & big block Chevy v8s as well as the newer LS style engines.

If I recall the S10 series trucks ran the NV3500 manual transmission. This is the same manual transmission that is installed in the C/K 1500 series trucks. The final answer is yes it will work if it is the NV3500.
