A tire does not fill any faster with the weight of the car on it. Air is pressurized and fills the tire with the same speed whether it is on or off of the car.
On a 2000 Ford Ranger : On a manual transmission , underneath the vehicle , on the transmission will be a fill plug . With the vehicle level the fill plug is cleaned and removed to check the fluid level ( the fluid level should be at the bottom of the fill plug hole )

Since the subject vehicle is probably a 4x4 there are two 'gear box' possibilities. If you mean Transmission, there is a right side plug that can be removed (from under the vehicle) with a 3/8 square drive. If you mean Transfer case, there is a fill plug and a level plug, remove both of these, one with 3/8 square drive and one a 10mm wrench.
