Yes Alice does lose her powers. On Resident Evil: Afterlife, Alice is injected with a serum by Wesker that eliminates the t-cells in her body, taking away her powers. Alice is still tough though, and glad that she is human again.
Yes Jill Valentine is still alive within the Resident Evil film series. In the novelization of Resident Evil: Extinction it is explained that Jill eventually leads her own group of survivors after proving to be a great asset. In Resident Evil: Afterlife the current movie, Jill is in the final scene, shown to be bad because she is being controlled by Wesker, like in the Resident Evil 5 game.
There are infact plans for a sequel to RE: Afterlife. Milla Jovovich (Alice) said in an interview to New York Magazine that there are plans for a 5th RE film and that it will feature characters and ideas that will be the result of feedback from fans. In a separate interview, Ali Larter (Claire) and Wentworth Miller (Chris) said that they would repirse their roles if asked. Sienna Guillory reprised her roll as Jill in the after-credits scene of RE: Afterlife, so it is largely assumed that she will be the antagonist of the 5th film, due to the fact that she is still under the control of Umbrella in the film continuity. Writer/Director Paul Anderson said that he would also like for the character Leon S. Kennedy to make an appearance as well.

Its rumored that Anderson signed for 2 more movies so Afterlife will be released September 10th 2010 and then he should start to work on the 5th one but after that I believe the series is getting a reboot and Anderson shouldn't be working on it anymore
