Eosin methylene blue agar (EMB) is a selective and differential medium used to isolate fecal coliforms. Eosin Y and methylene blue are pH indicator dyes which combine to form a dark purple precipitate at low pH; they also serve to inhibit the growth of most Gram positive organisms. Sucrose and lactose serve as fermentable carbohydrate sources which encourage the growth of fecal coliforms and provide a means of differentiating them. If the pH is changed (even at the beginning) the test might be ''read" incorrectly and the improper medication will be given. Always follow instructions for this reason.

Changing the starting pH will alter a mediums specificity or sensitivity on an EMB agar. This is because equilibrium is no longer met.

Would a change in starting pH alter the mediums sensitivity or specificity Explain?

Yes, it would. Most microbes grow best at what is called an optimum temperature and pH. If the pH is not at the optimum, the microbe may grow slowly or perhaps not at all.
