It couldn't be more than an hour on a plane, and probably more like half an hour. Most people don't fly from New York City to Southampton. Most people drive, take the bus (the Hampton Jitney) or the train (the LIRR, the Long Island Railroad).

Long Island is about 100 miles long, and Southampton is relatively close to the eastern end of it. Manhattan is just across the East River from the western edge of Long Island (Queens and Brooklyn). And since JFK and LaGuardia airport (2 of the 3 airports that service the NYC area, besides Newark) are actually located ON western Long Island (in Queens)...the distance by plane is less than 100 miles. Probably more like 70 miles.

I've flown from New York City to central Pennsylvania, which is over twice the distance from New York City to Southampton. And that flight took about an hour. So the flight from New York City to Southampton would be less than an hour. Again, probably more like half an hour.
