Exactly the same thing that would cause an ignition miss in any other engine.
The question becomes, "how do you know that it's an ignition miss as opposed to a possible burned valve?"
If you're certain it's caused by ignition, check the plugs, cap, wires and the ignition pickup assembly.

The wires are different - HEI ignition wires won't work in a standard distributor - they all go in the same relative places though.

The 327 cubic inch Chevrolet's distributor had points and condenser and not an ignition module. If you have an ignition module then you have a HEI distributor that was changed out.
