On a 2001 Ford E350 with a 7.3L diesel engine, the fuel filter is located on the driver's side frame rail, just before the fuel tank. It's typically mounted in a vertical position and can be accessed from under the vehicle. Make sure to relieve the fuel system pressure before attempting to change the filter to avoid spills.

If it is a Diesel Powered Van it is on the Drivers side Framerail directly below the door, next to the transmission. Not sure on a gasoline powered van, but its probably in the fuel tank.
The diesel engine that would be in a 2008 Ford E-350 van would be the 6.4L Powerstroke Turbo Diesel V8. This Powerstroke engine got poor fuel economy with about 16mpg highway and 12mpg city.
