The death of the federalist party had a lot to do with the fact that all of their great leaders had died and no one new stepped up. President Madison was pretty much the last outstanding federalist and he was shot in a duel. That shot is said to be the mortally wounding shot to the federalists.

As for New England, it is all about the Hartford convention. There were there was some talk about secession, but it is mostly exaggerated by the history books. (they mostly talked about 2/3 vote in congress for war and tariffs)

How did political parties contribute to the death of Alexander Hamilton?

Even though Hamilton was a Federalist the "Federalist" wasn't considered a political party as we know it today. It was just one side in the debate on the foundation of the constitution. It did not lead to his death. His death was caused by a duel with Burr and it was more personal than political. Burr cheated in the duel. His gun had a hair trigger. This was dicovered a few years ago when the dueling set was X rayed by historians.
