A common answer is Pheidippides, however he was already dead - having run to Sparta and back to invite them to come to help fight off the Persians, and he died of exhaustion ten days before Marathon.
After the victory at Marathon, the Athenian army of 9,000 armoured warriors ran back to Athens to protect it as they had realised that the Persian fleet was rowing the Persian cavalry around to Athens to gallop up and seize the city whose gates would be opened by traitors. The 9,000 arrived back just in time to stop this.
So the person(s) were 9,000 Athenian soldiers who ran carrying their armour and weapons. Today's marathon runners get off lightly, not carrying the load or wearing sandals. They get off lightly in their pretence of matching the run.
Though considered a myth, Pheidippides ran from Marathon to spread the news of the victory against the Persians. That said, his epic destination was not Rome. It was Athens.

Athens' feelings towards Sparta could differ from person to person and most important from time period to time period. A messenger by the name of Pheidippides, although there is some contention as to whether this actually happened. Pheidippides WAS sent with a request for aid from Marathon to Sparta, which was a distance of 240 km (which only took him two days). The story relates that he traveled back to the battle was was then sent to Athens with news of victory. Then, after running only 40km, he collapsed dead after announcing the victory.
