It depends on what kind of lip piercing you're talking about. For a labret piercing, it'd seriously affect the reed and make it impossible to play. Plus it'd hurt like hell.
For a piercing on the side (snake bites, spider bites, a ring on the side) it would probably affect your embouchure, but it would likely still be possible to play.
If you get your lip pierced in the middle you wont be able to play at all, atleast not with the piercing in, and even then, after getting it done you are not suppose to take it out for 4-6 weeks to let it heal. if its on the sides of your lips you can play the day you get it pierced
Give the piercing at least 3 or 4 days to settle down and get over any swelling. Use new reeds when you play for the first while to avoid the possibility of bacteria building on the used reeds or the chance of cross contamination. If you find your lip is sore after doing your exercises then give it a couple more days to settle. Practice practice practice.

It varies from person to person and depends on how long you have had the piercing. If it's under a year and a half (minimum preferably you should wait 2 years) then you should not leave it out for more than a few minutes.
