Cylinder misfire can generally occur not only to worn ignition components like the spark plug or wire, though can also be attributed to a faulty fuel injector or a gummed injector. Certain events in a coil-on-plug(COP) design(no wires) if you find your spark plug is known-good or new and gaped properly the coil itself may be faulty/shorted. Looking further, you may have to get further diagnostics to ensure your computer system is sending proper information to the coil(all sensors working in sync).
