Theory #1 Moisture can be sucked into the well if you have a blown head gasket near that plug and could cause rust or what appears to be rust from the burnt antifreeze after boiling!...
Solution: remove the plug wire from the sparkplug (before you start engine) so that you can see the bottom of the spark-plug with a flashlight, Remove all water, then start the engine. Look for bubbles...it may be several minutes until engine warms and yes your engine will sound rough of course, but if you see moisture appear Magically its a small leak .. seek the professionals!
Theory #2 Moisture from condensation could be drawn to that cylinder through the spark-plug wire via a cracked wire or spark-plug boot.

You must remove the plastic resonator box that says "Vortec 4200" on the top of the motor to see where they are. After you do that you will see 6 spots where you must unbolt the coil packs and lift them off the spark plugs. The spark plugs are down inside those holes.
