Differential media is able to distinguish different species or types of microorganisms based on an observable change in the colonies or in the medium. This type of medium contains specific nutrients or indicators that allow for differentiation between microorganisms based on their metabolic capabilities or other physiological characteristics.
Tryptone broth is neither differential nor selective media. It is a nutrient-rich medium used for the growth of a wide range of microorganisms.
Litmus milk contains several components such as; lactose (milk sugar) casein (milk protein) and Litmus ( Ph indicator) that can be metabolized. Thats why it is considered a good differential medium
The use of a medium that is both selective and differential can be justified in a few ways. They allowe the growth of organisms that can be of interest. The media can also be quite useful diagnostically in medical settings.
An example of a differential medium is MacConkey agar, which is used to isolate and differentiate gram-negative bacteria, particularly enteric bacteria. It contains bile salts and crystal violet, which inhibit the growth of gram-positive organisms, while lactose and a pH indicator allow for the differentiation of lactose fermenters (which produce pink colonies) from non-fermenters (which remain colorless). This selective and differential property makes it useful for identifying specific bacterial groups based on their metabolic characteristics.
Luria-Bertani (LB) broth is primarily considered a rich medium rather than a selective or differential medium. It supports the growth of a wide variety of bacteria, particularly Escherichia coli, but does not contain specific agents to inhibit or promote the growth of particular organisms. Selective media contain inhibitors to suppress unwanted organisms, while differential media contain indicators that allow for the distinction between different types of bacteria based on their biochemical properties.
EMB is an undefined selective/differential medium. It contains aniline dyes (methylene blue and eosin), which inhibit the growth of Gram-positive bacteria selecting for Gram-negative bacteria. EMB also contains lactose which makes the media differential based on an organisms ability to ferment lactose
